In 1833, the town of Lancaster was formed from part of the town of Clarence. The town was given the name Lancaster after Lancaster, Massachusetts, but the inspiration for this choice is unknown. Lancaster has the oldest stone building in Erie County, the Warren Hull House. In 1992, it was entered on the National Register of Historic Places. The Gipple Cabin, the oldest wooden building in Erie County, is also located in the town. In 1913 and 1914, the Lancaster Malleables, who participated in the New York Pro Football League, took home the league title!

Early in the 1990s, the town saw a period of fast growth, with a lot of construction taking place in its southern and eastern regions. In terms of both student enrollment and campus size, Lancaster High School is the biggest school in Erie County. In the village of Lancaster, some of the major community events are: The Taste of Lancaster, a restaurant festival and charity event, and the Fourth of July Parade.
